چکیده

When murine L1210 leukemia cells growing in suspension culture are deprived of tryptophan, there is a rapid fall in DNA and histone synthesis and a less rapid fall in whole cellular protein and non-histone nuclear protein synthesis. Initially, the S phase of the cell cycle becomes prolonged, but subsequently the cells become defective in their ability to reinitiate S phase. By 24 hr complete growth arrest has occurred, at which point about 60% of the cells have a GI DNA content and 32% have a late-S-phase DNA content. Growth arrest occurs at a cell density about 1.8 times the starting density. Readdition of tryptophan at this time pro duces resumed incorporation of thymidine-3H followed by

Effect of rutin on oxidative DNA damage in PC12 neurons cultured in nutrients deprivation condition

Objective(s): Rutin is a flavonoid with potent antioxidant property, which exhibited cytoprotective effects in several models of neuronal injury. This work aimed to examine whether rutin can protect neurons against oxidative DNA damage caused by serum/glucose deprivation (SGD) as an in vitro model of neurodegeneration and ischemia. Materials and Metho...

Can social instability, food deprivation and food inequality accelerate neuronal aging?

Based on both animal and human studies, inequality in food intake and social instability has adverse effects on the health of individuals and the community. However, it is not known whether social instability, food deprivation and food inequality affect neuronal death and premature aging in young animals. To address this question, the effects of these adverse situations, histopathological chang...

Evaluating the Viability of Adipose Tissue-derived Mesenchymal Stem Cells in Diabetic Culture Media Model

Introduction: In diabetes mellitus, stem cells are exposed to inappropriate conditions such as an increase of glucose in extracellular space, oxidative stress, and deprivation from growth factors (following ischemia). This study was aimed to evaluate the effect of these conditions on stem cells. Methods: Stem cells were isolated from subcutaneous adipose tissue of rats. The cells were maintaine...

Nimodipine Protects PC12 Cells against Oxygen-Glucose Deprivation

The protective effect of a L-type calcium channel blocker, nimodipine, on cell injury induced by oxygen-glucose deprivation (OGD) in PC12 cells was investigated. PC12 cells were exposed to in-vitro oxygen-glucose deprivation (30 minutes and 60 minutes respectively) in the presence or absence of nimodipine (10mM/L) in three different time schedules (pre-24h, pre-3h and concurrently). Cellular vi...

Effects of combinations of drugs having different modes of action at the ribonucleotide reductase site on growth of L1210 cells in culture.

Combinations of inhibitors directed at the individual components of ribonucleotide reductase were studied for their effects on L1210 cell growth in culture. The combinations included pyrozoloimidazole (IMPY) plus deoxyadenosine and hydroxyurea plus deoxyadenosine. Modulators were utilized to potentiate the effects of hydroxyurea, IMPY, or deoxyadenosine. Desferal was used to modulate the activi...
